■ 주요 연구내용
- | 폐 (Lung)는 외부와 접하고 있는 부분이기 때문에 공기나 혈관을 통한 pathogen, chemical에 의해서 계속적인 손상을 받고 있습니다. 특히 최근의 코로나 사태 (Covid-19 Pandemic)와 가습기 살균제로 인한 급성 폐 섬유화 이슈에서 알 수 있듯이 폐는 바이러스에 의한 1차적 공격의 대상 임은 물론 우리의 일상 과도 밀접하게 연관되어 있는 기관입니다. 저희 연구실은 손상 받은 폐가 어떻게 복구되는지 조직재생 (Tissue Regeneration)에 관한 연구를 하고 있습니다. |
- | 세포 손상 이후 면역 시스템 (Immune system) 의 변화로 인환 염증반응 (Inflammation)에 의해서 어떻게 폐에 존재하고 있는 상피 줄기세포 (Tissue resident epithelial stem cells)의 활성과 분화가소설 ( cell fate decision/celluar plasticity)이 변화하는지를 연구합니다. |
- | 또한, 줄기세포의 미세환경 (microenvironment)를 구성하고 있는 neighbouring niches (e.g. stromal cells, endothelialcells etc)들이 어떻게 변화하고 조직재생에 관여하는지를 살펴 보고자 합니다. |
- | 궁극적으로 이러한 재생과정이 만성 폐질환 (Chronic lung diseases) 특히 폐암 (Lung cancer)과 폐섬유화 (Pulmonaryfibrosis)와 연관이 있는지를 연구하고자 합니다. |
■ 세부 연구내용
(1) in vivo Genetically Engineered Mouse Models (GEMMs): 마우스 모델
: 본 연구실에서는 다양한 유전적
변이 마우스 모델을 사용하고 있습니다. 특히 stem cell의 behavior를 추적관찰하기 위하여 lineage-tracing mouse system을
가장 많이 사용하고 있습니다.
(2) ex vivo human/mouse 3D organoids model system: 오가노이드
모델
: 본 연구실에서는 폐조직에 상주하는 조직 내인성 성체줄기세포
기반의 오가노이드 모델을 활용하고 있습니다. 특히, 줄기세포와
면역세포, 섬유아세포의 공배양 시스템을 활용하여 다양한 세포들간의 상호작용을 연구하고 있습니다.
(3) Single cell multi-omics: 단일세포 멀티오믹스 분석
: 본 연구실에서는 조직 재생 과정에서 줄기세포 및 주변
미세환경의 유전적/후성유전적 변화를 연구하기 위하여 단일세포 기반의 멀티오믹스 분석을 활발하게 분석하고
있습니다.
